چکیده

The transcription of the T5 first-step transfer genes A2 and A3 has been shown to proceed from right to left (i.e., from A3 to A2) on the T5 DNA molecule.

Location of single-strand interruptions in the DNA of bacteriophage T5.

The positions of three single-strand interruptions in the DNA of phage T5(+) have been located by electron microscopy. All three interruptions were found in the same strand. Uneven base composition along the molecule is indicated by the preferential melting of certain regions. The data suggest a model according to which (1) the first-step-transfer DNA section is separated by a single-strand int...

Polarized injection of the bacteriophage T5 chromosome.

Examination of the first-step-transfer DNA of T5ris mutants which carry new EcoRI sites showed that the left end of the chromosome is injected first.

Evidence for heterogeneity in populations of T5 bacteriophage.

Each T5 stock contains a population of particular phages that, just after adsorption onto the host bacteria. release their entire chromosome outside the bacterial membrane in a place where it is sensitive to bacterial enzymes. This release takes place before the sensitization step to deprivation of calcium and before the transfer of the first-step DNA fragment. Secondarily, this released DNA is...

Characterization of an unusual bipolar helicase encoded by bacteriophage T5

Bacteriophage T5 has a 120 kb double-stranded linear DNA genome encoding most of the genes required for its own replication. This lytic bacteriophage has a burst size of ∼500 new phage particles per infected cell, demonstrating that it is able to turn each infected bacterium into a highly efficient DNA manufacturing machine. To begin to understand DNA replication in this prodigious bacteriophag...

Fluorescence changes of a membrane-bound dye during bacteriophage T5 infection of Escherichia coli.

The fluorescence intensity of membrane-bound N-phenyl-1-naphthylamine increases dramatically when T5 bacteriophage infect colicin Ib plasmid-containing hosts. This dramatic increase is not seen during normal infections or in infections wherein either the plasmid or the phage contain mutations which allow productive infection to occur. Two smaller increases in fluorescence intensity are seen, ho...
